A long-standing question in anesthesia is that of the molecular mechanism. Do anesthetics target proteins or change membrane properties, or both?  Using Fluorescence Cross-correlation Spectroscopy, Fluorescence Spectroscopy and Atomic Force Microscopy we have investigated the effect of the anesthetics halothane and ethanol on the structure and dynamics of phospholipid bilayers. We propose that an anesthetic-induced increased area per lipid drives local chain disorder thus promoting local phase change. We also examined the effect of halothane in promoting vesicle fusion.
